Single nucleotide base substitution (Glu? Arg) at 1 of cleavage sites where factor V is cleaved by activated protein C; results in abnormal factor V. Mutation may be involved in 50% of unexplained cases of venous thrombosis and occurs in 95% of activated protein C resistance. Positive result should be viewed as risk for thrombosis (not absolute marker).
